CENP-A chromatin prevents replication stress at centromeres to avoid structural aneuploidy

چکیده
Significance CENP-A, the histone H3 variant that forms a unique centromeric chromatin, is essential for faithful chromosome segregation during mitosis. Inability to connect centromere mitotic spindle causes aneuploidy, hallmark of many cancers. In addition missegregation, fusions at (peri)centromeres are prevalent in cancers, but how such rearrangements arise remains unclear. Here, we identified role CENP-A maintaining integrity centromere-associated repetitive sequences by ensuring their effective replication human cells. absence generation DNA–RNA hybrids due transcription–replication conflicts delayed DNA replication, breakage, recombination, and translocations centromeres. Centromeres thus possess special mechanism facilitate suppress translocations.

CENP-C recruits M18BP1 to centromeres to promote CENP-A chromatin assembly
Eukaryotic chromosomes segregate by attaching to microtubules of the mitotic spindle through a chromosomal microtubule binding site called the kinetochore. Kinetochores assemble on a specialized chromosomal locus termed the centromere, which is characterized by the replacement of histone H3 in centromeric nucleosomes with the essential histone H3 variant CENP-A (centromere protein A). Understan...
متن کاملHeterochromatin and RNAi are required to establish CENP-A chromatin at centromeres.
Heterochromatin is defined by distinct posttranslational modifications on histones, such as methylation of histone H3 at lysine 9 (H3K9), which allows heterochromatin protein 1 (HP1)-related chromodomain proteins to bind. Heterochromatin is frequently found near CENP-A chromatin, which is the key determinant of kinetochore assembly. We have discovered that the RNA interference (RNAi)-directed h...
متن کاملIdentification of Noncoding Transcripts from within CENP-A Chromatin at Fission Yeast Centromeres*
The histone H3 variant CENP-A is the most favored candidate for an epigenetic mark that specifies the centromere. In fission yeast, adjacent heterochromatin can direct CENP-A(Cnp1) chromatin establishment, but the underlying features governing where CENP-A(Cnp1) chromatin assembles are unknown. We show that, in addition to centromeric regions, a low level of CENP-A(Cnp1) associates with gene pr...
متن کاملNew insights into how chromatin remodellers direct CENP-A to centromeres.
Centromeric nucleosomes contain the unique histone H3 variant centromere protein A (CENP-A). How CENP-A is specifically loaded onto centromeres and maintained is an active area of investigation and several SNF2 family chromatin remodellers have been implicated in this process. In this issue of The EMBO Journal, Gkikopolus et al (2011) demonstrate a novel function for the SWI/SNF complex in remo...
متن کاملPolo-like Kinase 1 Licenses CENP-A Deposition at Centromeres
To ensure the stable transmission of the genome during vertebrate cell division, the mitotic spindle must attach to a single locus on each chromosome, termed the centromere. The fundamental requirement for faithful centromere inheritance is the controlled deposition of the centromere-specifying histone, CENP-A. However, the regulatory mechanisms that ensure the precise control of CENP-A deposit...
